Many years ago, diphtheria toxin (DT) showed antitumor activity in mice and in humans, but it was unclear whether this depended on the toxicity of the molecule only or on its strong inflammatory-immunological property as well. (Buzzi S., Cancer Res. 1982 May;42(5):2054-8). The same researchers, to deal with this open question, planned to treat a group of cancer patients with cross-reacting material 197 (CRM197).

An oncolytic adenovirus which calls macrophages in action

A report on the creation of a virus obtained genetically modifying a common adenovirus which could constitute a therapy against cancer has appeared recently in the scientific news. This virus would selectively infect cancer cells and force them to express a protein which calls for the intervention of macrophages, additionally stimulating them to multiply. This strategy would therefore help the body mounting a strong immune response against cancer cells.

The concept of Immunotherapy is based on the body's natural defense system, which protects us against a variety of diseases. Although we are less aware of it, the immune system also works to aid our recovery from many illnesses. For many years, physicians believed that the immune system was effective only in combating infectious diseases caused by such invading agents as bacteria and viruses. More recently, we have learned that the immune system may play a central role in protecting the body against cancer and in combating cancer that has already developed. This latter role is not well understood, but there is evidence that in many cancer patients the immune system slows down the growth and spread of tumors.

Standard cancer therapies

At present therapeutic protocols are much articulated. Usually they use conbinations of surgery, radiotherapy and chemotherapy. Target therapy is currently the new frontier and companies are developing molecules "specifically" (almost, in many cases..) directed against molecular features of tumors.Nanotech is considered  a technical step forward.

About conventional protocols one can find plenty of information at www.cancer.gov or at www.cancer.org, to mention only the most famous sites. Novel drugs are introduced into the market after having passed clinical trials, regulated by the FDA in the USA or the EMEA in the European Union.

 
Why doctors don't use alternative remedies

In a word, fear. And, it may surprise you but they can't anyway.

The small percentage of MDs who do know about them sincerely wish they could, but they put themselves, and their clinics/hospitals, at legal and professional risk unless they use radiation, surgery and drugs alone. Not a happy message, but this is the way of the world and we must face it. Medical politics at the federal and state level rules our doctors. All MDs fear powerful government regulatory agencies, especially the federal bureaus.The pharmaceutical drug orientated FDA has a decades-long record of not approving, or even looking at, anything alternative - period.Again, this is the way of the world and you must face it.

In the review paper "Dichloroacetate (DCA) as a potent..." appeared in the Online first session, in September issue of BJP, Prof. Michelakis most interestingly underlines the importance of targeting the peculiar "aerobic glycolytic" metabolic status of tumors. He suggests that the "Warburg effect", as the phenomenon is more commonly known in the literature, can be modulated by unlocking a mitochondrial impairment in cancer cells and that this could be a very promising strategy to fight cancer.

Can metformin boost DCA?

By itself, metformin was ineffective in treating tumors.
